Preclinical Data Supports Continued Advancement of SZN-1326 and SZN-043 Toward First-in-Human Clinical Studies in 2022
SOUTH SAN FRANCISCO, Calif., May 24, 2021 (GLOBE NEWSWIRE) — Surrozen Inc., a biotechnology company discovering and developing drug candidates to selectively modulate the Wnt pathway, announced today that data from preclinical studies of its two lead Wnt-modulating antibody programs were presented in six oral and poster presentations during Digestive Disease Week 2021.
Of the six presentations, two discussed data from studies of SZN-1326, a bi-specific full-length human antibody targeting Fzd5/8 and Lrp6 receptors in development for the treatment of inflammatory bowel disease (IBD). Four presentations discussed data from studies of SZN-043, a hepatocyte-targeted R-spondin mimetic for severe alcoholic hepatitis. SZN-1326 Presentations
In an oral presentation titled, “SZN-1326, a Wnt signal activator, improved epithelial healing and ameliorated colitis in an acute DSS model,” data showed SZN-1326’s ability to promote mucosal healing and reduce inflammation. As part of the study, mice were subjected to dextran sulfate sodium (DSS) and developed severe colitis. Treatment with SZN-1326 resulted in improved body weight, repaired damaged colon epithelium, and decreased serum and colon inflammatory cytokine markers without causing hyperplasia in healed epithelium or small intestine.
In a poster presentation titled, “SZN-1326 Promotes Colonic Mucosal Healing in an Acute Injury Model of IBD by Expanding the Progenitor Pool and Accelerating Differentiation,” data showed mechanisms underlying the molecule’s ability to promote mucosal healing by inducing stem and progenitor cell expansion that ultimately accelerates epithelial repair and reduces inflammation.
SZN-043 Presentations
In an oral presentation titled, “SZN-043, a Hepatocyte-targeted R-spondin Mimetic, Stimulates Hepatocyte Proliferation in an Acute Alcoholic Hepatitis Model,” the data showed the potential of SZN-043 to stimulate hepatocyte-specific cell regeneration, to activate the Wnt signaling pathway and to improve hepatic function in aging mice after a prolonged chronic binge ethanol treatment.
In an oral presentation titled, “SZN-043, a Hepatocyte-targeted R-spondin Mimetic, Induces Hepatocyte Proliferation in an Acute Acetaminophen-induced Liver Injury Model,” results demonstrated SZN-043 can induce hepatocyte-specific regeneration in response to an acetaminophen overdose reducing the area of necrosis.
In a poster presentation titled, “SZN-043, a Hepatocyte-Targeted R-spondin Mimetic, Promotes Transient Hepatocyte Proliferation and Zonal Gene Expression Changes in Mice,” results demonstrated the program’s ability to target hepatocytes and promote their proliferation, suggesting its utility as a regenerative therapy for liver diseases.
In a poster presentation titled, “Pharmacokinetics, Pharmacodynamics, and Toxicology of SZN-043, a Hepatocyte-targeted Wnt-potentiator, in Non-human Primates,” data demonstrated that non-human primates could be dosed up to 125 mg/kg twice weekly for 2 weeks without adverse effects. SZN-043 showed target-mediated pharmacology and the pharmacokinetics of SZN-043 were consistent with that of an IgG-based molecule binding to the hepatocyte membrane protein ASGR1.
