Insilico Medicine Announces the Nomination of Two Preclinical Candidates for PHD2, 12 Months After Program Initiation
NEW YORK and SHANGHAI, Jan. 5, 2022 /PRNewswire/ — Insilico Medicine, a global clinical-stage biotechnology company specializing in the applications of end-to-end artificial intelligence (AI) for drug discovery and development, today announced that the company has nominated preclinical candidates (PCC) for ISM012-077 and ISM012-042 for the treatment of anemia of chronic kidney disease (CKD) and inflammatory bowel disease (IBD), respectively.
ISM012-077 and ISM012-042 are small-molecule prolyl hydroxylase domain 2 (PHD2) inhibitors with novel scaffolds and distinct binding modes, demonstrated by a co-crystal structure. ISM012-077 is a potential best-in-class PHD2 inhibitor for anemia of CKD treatment that promotes erythropoiesis through both erythropoietin (EPO) induction and improved iron utilization, while demonstrating a favorable safety profile in vitro and in vivo. ISM012-042 is potentially a first-in-class gut-restricted PHD2 inhibitor for IBD treatment that induces the expression of gut barrier protective genes.
